Abstract

Orlistat (Xenical) is a lipase inhibitor. It was designed to address the problem of obesity and obesity associated with obesity-related side effects. In this randomized, double-blind, parallel, placebo-controlled study, 120 men with obesity-related obesity-related side effects and 60 healthy non-obese controls participated in a 2-week washout period. All participants were instructed to take a 60-mg dose of Orlistat or a placebo (either Xenical or dummy) for 12 weeks. All subjects had at least a 2-week washout period. After 12 weeks, participants were assessed for the presence of side effects using the Xenical, placebo or Xenical (Xenical 120 mg) versus the placebo (or dummy) endpoint. The percentage of subjects who experienced side effects was compared between the two groups at 12 weeks, at each point in the washout period, and at every time point. The Xenical effect size was 0.43 (SE: 0.31) in the Xenical versus the placebo group, and 0.33 (SE: 0.23) in the Xenical versus the dummy group. In addition, no statistical differences were found in the Xenical and the placebo effects between groups. The Xenical effect size was 0.36 (SE: 0.23) in the Xenical versus the placebo group and 0.35 (SE: 0.23) in the Xenical versus the dummy group. A statistically significant positive effect was found in the Xenical versus the placebo effect size for all three groups (p<0.001).
